She demonstrated in 2006 against the administration of Buddhadeb Bhattacharya's coercive land acquisition and mistreatment of local farmers under the pretext of industrial development policies.

Following this, Mamata was prevented from going to the Singur protest against the Tata Nano project. After voicing her opposition to this in the West Bengal Assembly, Mamata Didi called for a 12-hour Bangla Bandh. Mamata began a historic 26-day hunger strike in Kolkata on December 4.

To address the matter, then-President APJ Abdul Kalam spoke with then-Prime Minister Manmohan Singh. On December 29, at midnight, Mamata ended her fast. As soon as she took office as Chief Minister, she gave the Singur farmers back 400 acres of land. The Left Front government of West Bengal donated 997 acres of land for the Tata Motors facility in Singur, but the Supreme Court ruled in 2016 that this was unlawful.
